The breakthrough in her rehabilitation at a Houston hospital was first reported by Politico.
MSNBC quotes Pia Carusone, Giffords' chief of staff, as confirming the report.
It is the first time since she was shot in the head Jan. 8 that there has been a public confirmation that she has spoken.
"It's huge and it's great," says C.J. Karamargin, Giffords' spokesperson, the Arizona Daily Star reports. "It's very big news and we are all very excited."
